Abbexa's Bacterial Total Protein Extraction Reagent contains a gentle and nonionic detergent formulation which can quickly and efficiently extract total protein from bacteria without mechanical disruption. This reagent provides easy operation with high yields of the proteins. It is provided as a 10 ml bottle which is suitable for 50-100 assays.

Directions for use |
- Pellet bacterial cells by centrifugation at 1000 × g for 5 minutes. Aspirate the supernatant from the pellet. Suspend the bacterial cells with PBS, centrifuge, and then discard the PBS by aspirating the supernatant.
- Suspend the cell pellet with precooled Bacterial Total Protein Extraction Reagent at a ratio of 1:10-50 (e.g. Suspend 100 µl of cell pellet with 1-5 ml of Extraction Reagent). Mix well by shaking for 30-60 minutes.
- If required, ultrasonicate at in 10-30 second bursts, for 4 bursts in total, avoiding foaming. Reduce the temperature of the cell pellet using an ice water sonicating bath.
- Centrifuge the lysate at 10,000 × g for 10 minutes at 4 °C.
- Carefully transfer the supernatant to another tube, taking care to not disturb the insoluble pellet, then place the tube on ice for future use in protein assays. If required, remove agglomerated proteins by centrifugation at 10,000 × g for 30 minutes at 4 °C, then carefully transfer the supernatant to another tube, then place the tube on ice for future use in protein assays.
