DAPI Staining Kit for examining cellular DNA in fluorescence microscopy.
DAPI (4',6-diamidino-2-phenylindole) is a cell-permeable fluorescent stain that binds to the AT-rich regions in double-stranded DNA in both live and dead cells. The DAPI-DNA complex exhibits a light blue flourescence color with excitation light 364 nm and emission light 454 nm.
Kit Components: - DAPI Chromogen (1 mg/ml): 100 µl
- Dilution Buffer: 4 × 25 ml

Directions for use |
For double or triple fluorescence staining in immunofluorescence tests, DAPI staining should be carried out after all fluorescent dye-conjugated antibodies have been incubated. Suggested Procedure: - Tissue staining: Add 10-20 µl of DAPI Chromagen to 10 ml Dilution Buffer and mix to prepare the diluted DAPI solution. Incubate the tissue with diluted DAPI solution for 15-30 min, then wash with PBS/TBS 3 times. Cover the sample with a cover glass and observe under a microscope.
- Cell culture staining: Add 1-5 µl of DAPI Chromagen to 10 ml Dilution Buffer and mix to prepare the diluted DAPI solution. Incubate cells with diluted DAPI solution for 15-30 min at 30 °C. Cover the sample with a cover glass and observe under a microscope.
