Hoechst 33342/PI Double Staining Kit contains the Hoechst 33342 and propidium iodide stains for use in double fluorescence staining. Hoechst 33342 (Bisbenzimide) is a cell-permeable blue fluorescent stain that binds to adenine-thymine-rich regions of DNA, increasing its density. Propidium iodide (PI) is a red fluorescent stain that binds to DNA by intercalating between the bases. PI is unable to enter living cells, and so it stains cells that have undergone apoptosis, necrosis or fixation.

Recommended dilutions	IF/ICC: 5 µg/ml (total dye concentration, 1:1 ratio of Hoechst 33342/PI). The recommended concentration for all applications is 1- 5 µg/ml. Optimal dilutions/concentrations should be determined by the end user.
Storage	Store the Hoechst 33342 Chromogen and Propidium Iodide Chromogen at -20 °C, and the dilution buffer at 4 °C. Avoid exposure to light.

Directions for use	For double or triple fluorescence staining in immunofluorescence tests, Hoechst 33342/PI staining is the last step after incubating the antibodies. For cultured cells, add 10 µl of Hoechst 33342 Chromogen and 10 µl of Propidium Iodide Chromogen to 2 ml Dilution Buffer in the same tube. Mix thoroughly (total dye concentration is 5 μg/ml). Incubate for approximately 5 minutes in the dark at 30 °C, then wash with PBS/TBS 3 times, 3 minutes each time.
