Biological Activity Carcinoembryonic Antigens (CEAs) are members of the CD66 cluster of differentiation. They are glycophosphatidylinositol (GPI)-anchored intercellular molecules that are upregulated in many cancers, such as colon, breast and lung. Galectin 4 (GAL4) has been identified as an interactor of CEA, thus a binding ELISA assay was conducted to detect the interaction of recombinant human CEA and recombinant human GAL4. Briefly, CEA was diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100 µl were then transferred to GAL4-coated microplate wells and incubated for 2 h at 37°C. Wells were washed with PBST and incubated for 1 h with anti-CEA polyclonal antibody, then aspirated and washed 3 times. After incubation with HRP-conjugated secondary antibody, wells were aspirated and washed 3 times. TMB substrate solution was added and wells were incubated for 15-25 minutes at 37 °C. Finally, 50 µl stop solution was added to the wells and the absorbance was read at 450 nm immediately. The binding activity of CEA and GAL4 is shown in Figure 4.
