Biological Activity | Fatty Acid Binding Protein 4, Adipocyte (FABP4) encodes the fatty acid binding protein found in adipocytes. Fatty acid binding proteins are a family of small, highly conserved, cytoplasmic proteins that bind long-chain fatty acids and other hydrophobic ligands. FABPs may be involved in fatty acid uptake, transport, and metabolism. Actin Beta (ACTB) has been identified as an interactor of FABP4, thus a binding ELISA assay was conducted to detect the interaction of recombinant human FABP4 and recombinant human ACTB. Briefly, FABP4 was diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100 μl were then transferred to ACTB-coated microplate wells and incubated for 2 h at 37°C. Wells were washed with PBST and incubated for 1 h with anti-FABP4 polyclonal antibody, then aspirated and washed 3 times. After incubation with HRP-conjugated secondary antibody, wells were aspirated and washed 3 times. TMB substrate solution was added and wells were incubated for 15-25 minutes at 37 °C. Finally, 50 μl stop solution was added to the wells and the absorbance was read at 450 nm immediately. The binding activity of FABP4 and ACTB is shown in Figure 4. |
