| Introduction | Hyaluronidase 1 (HYAL1) is an enzyme that hydrolyses 1,4 linkages between N-acetylglucosamine (GlcNAc) and glucuronic acid (GlcA) residues in hyaluronan, a glycosaminoglycan found in extracellular matrix. HYAL1 is expressed in the lysosomes of various tissues and is also secreted in plasma. The catalytic activity of HYAL1 is carried out by an aspartate and a glutamate residue in close proximity to each other, but also requires a distant tyrosine and asparagine residue to ensure correct binding to hyaluronan. Overexpression of HYAL1 occurs in some metastatic cancers, while mutations in the gene that reduce its catalytic activity can cause mucopolysaccharidosis IX, resulting in short stature and high serum hyaluronic acid concentrations. |
