Biological Activity | A proliferation assay was conducted to detect the bioactivity of recombinant human IFNb using A549 cells. Briefly, A549 cells were seeded into triplicate wells in 96-well plates at a density of 2,000 cells/well and allowed to attach with serum-free medium overnight, then the medium was replaced with 5% serum standard DMEM prior to the addition of various concentrations of IFNb. After incubation for 96 h, cells were observed by inverted microscope (Figure 3) and cell proliferation was measured by Cell Counting Kit-8 (CCK-8) assay (Figure 4). Briefly, 10 µl of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 °C. The ED50 of recombinant human IFNb is 6.4 µg/ml. |
