Introduction | Neurofilament Light Polypeptide (NEFL) is one of five proteins that commonly form filaments in neuronal cells, with the other four being Neurofilament Medium and Heavy Polypeptide, as well as either peripherin or internexin. Together, these form neurofilaments, which form part of the neuronal cytoskeleton. NEFL and NEFM are found in close proximity to each other on human chromosome 8 and share some homology, suggesting that one could be the product of an ancestral gene duplication of the other. NEFL found in plasma or cerebrospinal fluid is used as a biomarker of nerve damage in multiple sclerosis and Alzheimer’s. Mutations in NEFL are associated with Charcot-Marie-Tooth disease. |
