Introduction | Osteopontin (also known as secreted phosphoprotein 1, bone sialoprotein I, BSP1, gene name SPP1) is a protein that binds to bone and prevents the formation of additional hydroxyapatite. This is accomplished via the negative charge produced by the base amino acid sequence as well as a large number of phosphorylation modifications that allow it to bind to calcium. SPP1 also provides a surface for osteoclasts to bind to, allowing them to begin the process of bone resorption. Outside of the bone environment, osteopontin promotes the adhesion, migration, and survival of immune cells. Dysregulation of SPP1 is associated with rheumatoid arthritis, inflammatory bowel disease, Duchenne muscular dystrophy, and various cancers. |
