Non-Liposomal Transfection Reagent contains a non-liposomal formulation designed to transfect DNA and RNA into a wide variety of eukaryotic cell lines with high efficiency and low toxicity. Primary cells and other difficult-to-transfect cells can also be effectively transfected using this reagent. This reagent can be used in the presence of serum and antibiotics.

Note This product is for research use only.
Directions for use Plasmid DNA Transfection: (for a 24-well plate)

  1. Seed cells: plate 2-6 × 104 cells in each well so that there is 70-90% confluence at time of transfection.
  2. Incubate for 12-24 hours.
  3. Dilute 0.8 µg of plasmid with 50 µl of reduced serum Minimal Essential Medium (MEM).
  4. Add 1.6 µl of premixed Non-Liposomal Transfection Reagent and mix gently.
  5. Allow to stand for 15-20 minutes at room temperature.
  6. Add the solution to each well containing cells and medium. Incubate cells at 37 °C in a CO2 incubator.
  7. Change medium 4-6 hours post-transfection (optional). Incubate cells for 18-72 hours.

siRNA Transfection: (for a 24-well plate)

  1. Seed cells so that there is 50-70% confluence at time of transfection.
  2. Incubate for 12-24 hours.
  3. Dilute 50 pmol of siRNA with 50 µl of reduced serum Minimal Essential Medium (MEM).
  4. Add 2 µl of premixed Non-Liposomal Transfection Reagent and mix gently.
  5. Allow to stand for 15-20 minutes at room temperature.
  6. Add the solution to each well containing cells and medium. Incubate cells at 37 °C in a CO2 incubator.
  7. Change medium 4-6 hours post-transfection (optional). Incubate cells for 18-72 hours.
