Liposomal Transfection Reagent is a liposomal formulation designed for use with difficult-to-transfect cell lines, such as neural and tumor cell lines, with high efficiency and low toxicity. This reagent can be used in the presence of antibiotics. Medium that allows reduced serum supplementation is required.

Directions for use Plasmid DNA Transfection: (for a 24-well plate)

  1. Seed cells: plate 2-6 × 104 cells in each well so that there is 70-80% confluence at time of transfection.
  2. Incubate for 12-24 hours.
  3. Replace growth medium with serum-free medium that allows reduced serum supplementation before transfection.
  4. Dilute 0.4 µg of plasmid with 100 µl of medium that allows reduced serum supplementation.
  5. Add 1.5 µl of premixed Liposomal Transfection Reagent and mix gently.
  6. Allow to stand for 10-20 minutes at room temperature.
  7. Add the solution to each well containing cells and medium. Incubate cells at 37 °C in a CO2 incubator.
  8. Change medium 4-6 hours post-transfection. Incubate cells for 18-72 hours.
