SDS-PAGE analysis, with Coomassie blue staining, of expression tests to determine induction strategy, temperature and time for induction, and E. coli strain to use for protein production. The negative control is marked Ø. Briefly, bacteria were obtained by incubation at 37 °C. IPTG induction was carried out at 16 °C for 16 h or 37 °C for 4 h. Bacterial cells were collected by centrifugation and lysed in a buffer containing PBS, pH 7.5, 10% glycerol. After centrifugation, the soluble supernatnant fraction was collected (native protein extraction - NPE). The insoluble fraction was collected and solubilized with a denaturing buffer (8 M urea). After centrifugation, the supernatant was collected (denatured protein extraction - DPE). Based on the expression tests, the optimal conditions were: T7E strain, IPTG induction at 37 °C for 4 h, NPE.

Oxyntomodulin is a peptide hormone produced by the cells of small intestine during food consumption and responsible for the sense of satiation owing to its effect on the hypothalamic nuclei. The physiological role of oxyntomodulin is to decrease appetite for homeostatic weight regulation. Being an innate human protein oxyntomodulin produces minimum side effects in contrast to known drugs for obesity. Human Oxyntomodulin Protein is a recombinant protein expressed in E. coli.
