Directions for use |
- Prepare the 1X Cell Lysis Buffer by adding nuclease-free water to the 5X Cell Lysis Buffer at a ratio of 4:1 (e.g. add 20 ml of nuclease-free water to 5 ml of 5X Cell Lysis Buffer to prepare 25 ml of 1X Cell Lysis Buffer).
- Prepare the Working Luciferase Reaction Reagent by adding 5 ml of Luciferase Reaction Buffer to 1 vial of lyophilized Luciferase Reaction Substrate. Mix thoroughly and ensure that the lyophilized powder has completely dissolved. The Working Luciferase Reaction Reagent should be used within one day, or aliquoted and stored in the dark at -20°C (up to one month) or -70°C (up to one year).
- Wash cells twice with PBS and add 1X Cell Lysis Buffer. If using a well-plate, the table below shows the recommended volume of 1X Cell Lysis Buffer.

Centrifuge at 12,000 × g for 10 minutes at 2-8°C. Transfer the supernatant (containing the cell lysate) to a new tube. - To 20 µl of cell lysate, add 100 µl of Working Luciferase Reaction Reagent. Measure the luminescence signal in a luminometer immediately.
