TMB Substrate is a ready-to-use reagent for use in ELISA. It reacts with horseradish peroxidase (HRP) to produce a blue colour. The reaction can be terminated by adding acidic stop solution (sulphuric or hydrochloric acid), which produces a stable yellow reaction product with absorbance maxima at 450 nm.

Directions for use |
- Add 100 µl of TMB to each microplate well.
- Incubate in the dark at 37 °C for 20-30 min. The reaction can be terminated when the desired color develops.
- Add 100 µl of stop solution (0.3 M H2SO4 or 0.25 M HCl) to each well.
- Read the absorbance of each well at 450 nm within 15 minutes.
Notes: - 0.1% NaF can be used as the stop solution instead of 0.3 M H2SO4 or 0.25 M HCl; the absorbance of each well should be read at 650 nm if using 0.1% NaF.
- Avoid contamination with oxidizing reagents.
- This product should be used in the dark. Do not use in metal containers.
- High HRP concentrations may cause the TMB to turn a green or red color and may cause precipitation to occur. If this occurs, it is recommended to reduce the HRP concentration in the sample or reduce the incubation time.
