Introduction | Tumour Necrosis Factor Ligand Superfamily Member 10 (TNFSF10, also known as TNF-related apoptosis-inducing ligand, TRAIL, and CD253) is a protein that induces apoptosis via Death Receptors 4 and 5 (DR4 and DR5). When TRAIL binds to these receptors caspase-8 is activated, which cleaves caspases 3, 6, and 7, leading to apoptosis of the target cell; alternatively, TRAIL can bind to decoy receptors, which prevent it from signalling. TRAIL is typically expressed by activated immune cells to kill infected and tumorous cells; conversely, TRAIL can also be expressed by cancer cells to kill approaching immune cells. Therapeutic agents targeted to TRAIL and its receptors have been investigated, but none of them have been approved for use at this time. |
